Detection of mutations associated with artemisinin resistance in K13 gene of Plasmodium falciparum parasites

Objective: this study aims to develop a methodology for detecting mutations in the K13 gene and determine mutation frequencies clinical samples. Method: total DNAs were extracted blood samples collected from 50 patients failure with artemisinin treatmentin Central Highlands region. A fragment of was amplified, purified, sequenced by Sanger method. The sequences analysed using Bioedit aligned reference sequence mutations. Results: successfully amplified segment size 799 bp P. falciparum. After sequencing, there nucleotide substitution A>G at position 1740, leading changes amino acids C>Y respective 580. In patient samples, 41/50 (82%) showed C580Y mutation, 9/50 (18%) had no other mutation. Conclusion: authors have developed optimised procedure falciparum determined frequency gene.
